چکیده

Nowadays, the discovery of lignocellulolytic microorganisms that are better adapted to operational conditions while exhibiting the strong degrading activities is highly desired for successful lignocellulose biotransformation processes. In this study, microorganisms were isolated from lignocellulose-rich composting materials by selective methods. A screening of isolates known to have lignocellulolytic abilities was performed using several tests. Seven microorganisms showed ligninolytic potential and were subjected for further analysis according to their degrading activity. The fungus Pseudallescheriaangusta MF4 demonstrated high decolorization rates for three aromatic dyes: Poly R478, Poly S-119, and Remazol Brilliant Blue R. In addition, the fungus showed a high production rate of ligninolytic enzymes in the presence of inducers. This fungus achieved the highest values of growth after 21 days of incubation on sawdust without any additional nutrients. Owing to its proven ligninolytic activity and capability of growing on a lignocellulosic substrate, the application of this isolate could be of interest in different biotechnological applications, particularly in biological treatment of wood fibres in order to improve the production of woodbased composites.

Molecular variability of Pseudallescheria boydii, a neurotropic opportunist.

The sequences of the internal transcribed spacer (ITS) ribosomal DNA (rDNA) domain data obtained by restriction fragment length polymorphism analysis with 18S rDNA and fingerprinting (M13) for clinical and environmental strains of Pseudallescheria boydii (anamorph, Scedosporium apiospermum) were compared to those for related species of Pseudallescheria, Petriella, and Scedosporium. The infraspe...

Susceptibility and diversity in the therapy-refractory genus scedosporium.

Scedosporium species show decreased susceptibility to the majority of systemic antifungal drugs. Acquired resistance is likely to disseminate differentially with the mode of exchange of genetic material between lineages. Inter- and intraspecific diversities of Scedosporium species were analyzed for three partitions (rDNA internal transcribed spacer gene [ITS], partial β-tubulin gene, and amplif...

Molecular Identification and Susceptibility of Clinically Relevant Scedosporium spp. in China

As various new sibling species within the Scedosporium spp. have been described recently, this study was conducted to investigate distribution and antifungal susceptibility profiles of the different species of Scedosporium spp. in China. Twenty-one clinical strains of Scedosporium from China and two strains from Japan were reidentified by MLSA. The analysis included BT2, CAL, RPB, SOD, and ACT ...

Improvement of ligninolytic properties in the hyper lignin-degrading fungus Phanerochaete sordida YK-624 using a novel gene promoter.

We identified a highly expressed protein (BUNA2) by two-dimensional gel electrophoresis from the hyper lignin-degrading fungus Phanerochaete sordida YK-624 under wood-rotting conditions. Partial amino acid sequences of BUNA2 were determined by LC-MS/MS analysis, and BUNA2 gene (bee2) and promoter region were PCR-cloned and sequenced. The bee2 promoter was used to drive the expression of the man...
